Vetri Duraisamy was born in Chennai, India, to Saidai Duraisamy and his wife. His father is a former mayor of Chennai and a politician. He belongs to the Kongu Vellalar community, a sub-caste of the Vellalars, who are traditionally farmers and landowners. He has a brother and a sister.
He completed his schooling at St. Bede’s Anglo-Indian Higher Secondary School in Chennai, where he developed an interest in filmmaking, photography, and aviation.

Vetri went missing on February 4, 2024, after the car he was travelling in with his friend Gopinath and driver Tenzin plunged into the Sutlej river in Himachal Pradesh.
